乳脂肪およびその分別物のリパーゼ分解 OAK

微生物起源の市販リパーゼを用いて、天然のバターフレーバーを強化する目的で乳脂肪を分解した。 1)市販酵素であるパラターゼAとリパーゼFを比較すると、 リパーゼFの方が分解率が高く、フレーバーも良好であった。 また、酵素添加量を増やすことにより短時間で分解率を上げることができた。 2)リパーゼFでバターオイルを分解した壌合、水相量の違い(10%と20%)、水道水の使用および水酸化物添加による加水分解率への影響はほとんどなかった。 3)リパーゼ分解により遊離した脂肪酸としては11種が検出され、そのうち主なものはパルミチン酸、ステアリン酸およびオレイン酸であった。24hと48hの分解時間で比較すると、遊離した脂肪酸量には違いがあったが、その組成はほとんど変わらなかった。またショートニング希釈およびマドレーヌでの実用試験においても、分解時間によるフレーバーの違いは単に強度的なものだけであった。 4)乳脂肪から分別された低融点画分を同様にリパーゼで分解すると、分解率はバターオイルの場合と比べて多少低かったが、遊離した短・中鎖の脂肪酸組成には差がなかった。官能試験では、刺激臭が弱くなり風味的にマイルドで、またマドレーヌでの実用試験にいでもフレーバー改善効果が認められた。しかし、これはバターフレーバー成分が分別の過程で低融点性画分に多く移行したことによると判断された。

Selection of Salt-tolerant Yeast Strains Accumulating Glucosylceramide OAK

Debaryomyces hansenii NBRC 0855 accumulated high amounts of glucosylceramide relative to 25 strains of salt-tolerant yeasts in the genera Candida,Debaryomyces. Kodamaea,and Pichia. The constituents of glucosylceramide were principally 2-hydroxystearic acid as a fatty acid,9-methyl-trans-4,trans-8-sphingadienine as a sphingoid base and glucose as a hexose. Strain NBRC 0855 was able to produce glucosylceramide when cultured on actual eflluent from a pickle manufacturer.

Selection of lactic yeast producing glucosylceramide from cheese whey OAK

From 2150 isolates from raw milk and milk products, yeast strains were surveyed to produce glucosylceramide from cheese whey. Most of the 54 strains that had accumulated a detectable amount of glucosylceramide were identified as Kluyveromyces lactis var. lactis. The cells of K. lactis var. lactis strain M-11 derived from domestic raw milk accumulated glucosylceramide 2.5-fold higher than K. lactis var. lactis NBRC 1267, the reference strain selected from the culture collections. Strain M-16 of K. lactis var. lactis derived from the same origin was found to synthesize a considerable amount of steryl glucoside in addition to glucosylceramide. Simultaneous Production of Sphingolipids and Ethanol by Kluyveromyces thermotolerans OAK

Kluyveromyces thermotolerans strain NBRC 1674 was selected for the simultaneous production of sphingolipids and ethanol from beet molasses. The strain gradually synthesized ethanol with fermentation periods and attained a level slightly higher than that of the strain of Saccharomyces cerevisiae usually used for ethanol production. The sphingolipids accumulated in the cells were composed of almost equal amounts of free ceramides and glucosylceramides. The sphingoid bases and fatty acids of the two sphingolipids differed from each other and changed under aerobic and anaerobic growth conditions. Oxygen limitation may cause accumulation of sphinganine by inhibiting sphingolipid desaturases and enhance its proportion in both the sphingolipids.

無水乳脂肪の分別画分の性状と分別条件の検討 OAK

無水乳脂肪を最終温度23℃で1段階自然分別して、ヨウ素価27で軟化点41.1℃の結晶画分(HF1)を24%の収量で得た。次いで、液状画分(SF1)に対して2段階分別を行い、分別物の収量と性状に及ぼす初期温度、最終温度および撹拌速度の影響を検討した。初期温度18℃と30℃の条件で比較すると、後者では不定形の大きな結晶(50μm程度)のみが析出して濾過が存易であった。得られた結晶画分(HF2)は、固体脂指数が高く、また1 段階分別で得られたHF1に次ぐ高い軟化点を有していた。SFlを冷却すると、12.5℃で温度が下がらない「hold状態」になり、500μm程度の不定形結晶と小さい150μm程度の球形結晶が得られた。それより少し高い温度(13℃)に設定すると、小さい球形結晶(100μm程度)のみが大量に生じた。この場合、濾過が困難であったがHF2の収量は43%と高かった。ヨウ素価から判断すると、得られたHF2は12.5℃の条件と比べて融け易い性状であった。これは、中融点性のトリグリセリド種がより多く液状画分からHF2へと移行したことによると考えられた。撹拌速度については、速い条件(50rpm)でも遅い粂件 (28rpm)でも細かい結晶(50~100μm)が析出し、濾過効率の面から中間速度(43rpm)が適当であった。

赤ワインのアルコール発酵と乳酸発酵過程でのリスベラトロール関連物質量の変化 OAK

ワイン中の機能性物質のひとつであるリスベラトロールとその関連物質について,ワインの発酵過程での含量変化を分析した. (1) アルコール発酵3日目からのアルコール濃度の上昇とともにリスベラトロールは果皮から抽出され,発酵6日目でほぼ平衡に達した.その過程で,シス異性体の割合が増加し,発酵後半ではトランス体のみが減少していたことから,酵母由来のイソメラーゼ活性により発酵中にリスベラトロールの異性化が起きていることが示唆された. (2) 乳酸発酵過程において,リスベラトロールの増加とパイシードの減少が観察され,果皮から移行したパイシードが乳酸菌由来のβ-グルコシダーゼ活性によって加水分解されてリスベラトロールに変換されるものと推測された.During alcoholic fermentation trans-and cis-resveratrol were rapidly extracted from grape skin along with the rise in the alcoholic concentration and reached equilibrium six days after the start of maceration. Comparative Studies on lipid Analysis and Ultrastructure in Porcine and Southern Minke Whale (Balaenoptera Bonaerensis) Oocytes OAK

The present study was conducted to clarify the difference in the color of the cytoplasm in immature follicular oocytes from prepubertal and adult minke whales. The four lipid contents (triglyceride,total cholesterol phospholipids and non-esterified fatty acids) in vitrified immature oocytes from prepubertal and adult minke whales,and also in fresh and vitrified immature porcine oocytes,were measured. The lipid contents in vitrified-warmed minke whale oocytes were similarly high compared with those in vitrified-warmedp orcine oocytes. 大腸腺腫発症モデルマウスにおける食餌性ナガイモおよびナガイモ入り青汁の効果 OAK

食餌性ナガイモの大腸腺腫発症に与える効果を他のイモ類と比較しつつ,1,2-ジメチルヒドラジン投与マウスを用いて調べるとともに,ナガイモを利用した商品モデル(ナガイモ入り青汁)の効果についても検証した.その結果,ナガイモ以外のイモ類においても上記の実験動物モデルにおいて大腸腺腫抑制効果が観察されたが,腺腫数の平均はナガイモ群が一番低かった.また,商品モデルであるナガイモ入り青汁ついても腺腫抑制効果が観察された.The dietary effects of Chinese yam,nagaimo (Dioscorea opposita Thunb.),on 1,2-dimethylhydrazine (DMH)-induced aberrant crypt foci (ACF) formation in the large intestine of mice were investigated and compared with those of other potato species. Potato consumption was found to significantly suppress ACF formation in the large intestines of all mice treated with DMH. Suppression of ACF formation was most remarkable in the nagaimo group. Moreover, the commercial product (aojiru) containing nagaimo powder also suppressed ACF formation.
